Hyaluronic Acid
 

Hyaluronic Acid (hyaluronan, HA) is a naturally occurring sugar polymer of central biological importance to man. The magnitude of its importance has yet to be fully described. Some of what is known is:

1. The zona pellucida which is the protective, outer layer of the human egg (ovum) is composed of HA. The sperm must pass this barrier in order to fertilize the egg.

2. The developing embryo and fetus is bathed in HA. Scarless, in-utero surgery rests on the wound healing properties of HA.

3. The cement that holds cells together in the body (ground substance) is composed of HA, collagen and elastin. HA is responsible for the movement of water and other molecules in the body.

4. HA is present in the eyeball to keep it round and in the joint fluid as a lubricant.

5. HA, by virtue of the appearance of specific HA receptors, targets and accumulates in areas of damage, inflammation and infection. This is the basis of the HIT™ (HA Induced Targeting) drug delivery technology.

6. HA is the most potent naturally occurring humectant: It will bind 400 times its weight with water. The water binding capacity of HA is dependent on the size (molecular weight) of HA and on its purity.

7. It is present in skin to keep it moist. With age and exposure to sunlight the skin loses its ability to synthesize HA, reducing the moisture content of skin and consequently wrinkling. Indeed, at age 50, man has only 50% of the HA present at birth.

8. HA will deliver moisture to the skin and hold it there. It has been determined that HA will deliver water through the top (stratum corneum), epidermal and dermal layers of the skin. It has also been described and confirmed that in addition to water, HA will carry other molecules through these layers. The intradermal delivery of water and other materials by HA is also dependant on its molecular size and purity.

9. HA possesses wound healing and free radical scavenging properties. At higher molecular sizes it behaves like a viscoelastic (plastic) material. HA has been found to prevent arterial clogging (restenosis) after balloon angioplasty, scarring (adhesion formation) after surgery and is completely safe when infused intravenously in man.

10. HA is produced from animal sources (rooster combs) and by fermentation. It is generally available in two grades:

• Cosmetic Grade which contains many impurities and possesses a wide molecular size range and

• Medical grade which contains few impurities and is of a specific molecular size depending on its use. For example, high molecular weight HA is used in ocular surgery while lower molecular weight HA is used in joint injections.
